Project Name: Construction and evaluation of reverse genetics systems of human and porcine rotaviruses
Project Description: This project is supported by CVM Internal Research Competition (IRC). The goal of this project is to establish a plasmid-based reverse genetics system for generating infectious rotavirus particles. This system will be based on the recombinant vaccinia virus T7 RNA polymerase-driven procedure to supply artificial authentic viral mRNA which can be used for replication of double-stranded (ds) RNA and translation of rotavirus protein. The cDNA segments representing 11 genes of rotavirus will be cloned into individual plasmid vector pX8dT. The infection of rDIs-T7pol and transfection of recombinant plasmids containing the cDNAs of 11 rotavirus segments respectively will allow transcription of the positive-strand RNAs of rotavirus genome and translation of all rotavirus proteins. Our hypothesis is that the presentation of all rotavirus proteins and positive-strand RNA segments in cells can initiate the replication and packaging of rotavirus. Reverse genetics system will provide a tool for studies of pathogenesis and immunogenicity of rotaviruses and for development of effective and safe rotavirus vaccines for humans and animals.
